蒙娜丽莎的微笑1953年，坐落在美国马萨诸塞州、被誉为“没有男子的常青藤”的卫斯理女子学院，新来了位艺术史老师，名叫凯瑟琳·沃森(朱莉娅·罗伯茨 饰)，她是个美丽成熟的女人，刚刚从UCLA（美国加州大学洛杉矶分校)大学毕业，在大学里接受了自由改革思想，立志要成为一名杰出的教授。
50年代的美国虽然女性的地位渐渐受到重视，但在上层社会封建思想仍旧非常严重。在卫斯理这所著名的女子大学，学生们大都有着良好的家庭背景，从小接受过优秀的教育。但学院对学生的教育不是教他们如何获得自己感兴趣的学科知识，也不重视心理教育，而是把学生的成功与否定义为今后的婚姻，她们学习的目的无非是嫁一个好丈夫。以贝蒂(柯尔斯滕·邓斯特 饰)、琼(朱莉娅·斯泰尔斯 饰)和莉薇(玛吉·吉伦哈尔 饰)为代表的几个非常好动的女孩子，喜欢在班上卖弄风情，甚至用各种办法在课堂上向老师发起挑战。凯瑟琳没有像其他老师那样沿袭学校一贯的教学做法和风格，她不仅挑战学校的一些做法、规矩，而且鼓励学生发掘自己的兴趣，并且大胆去实践她们的想法。最终她以青春率直的作风，丰富的艺术史知识以及风趣热情的授课风格，赢得了学生们的尊敬和爱戴，被女学生称为“蒙娜丽莎”。
这样一位被学校的传统势力视做“异类”的女子能否成功达到她的目的，让她的学生们脱离束缚，找到自我？还是最终等待她的是那些传统观念，传统势力的猛烈“进攻”……
